Question Title What is the effective pixel size on the sample (i.e. what is the smallest spot that the camera sees)?
27.3 micrometers but we aggregate 3x3 pixels as minimum therefore the smallest spot is 3x3 pixels which is 82 x 82 micrometers.  For example:  Average hair thickness is 100 micrometers.
